I have hereditary dark circles, I know theres not much that could be done for hereditary type dark circles. But I would like to try something that can make it at least lighter. I have tried Garnier Under eye dark circle cream but didn’t work :( I have also tried potato juice which works as an instant solution. But I am looking for some over the counter remedies. Please suggest something that YOU HAVE TRIED and did work. (I am NOT looking for any concealor or make up product)

It is about optimal temperatures for human and fungal amylase:

Abstract:
This experiment revealed the optimal temperature needed for Human and fungal amylase to successfully do it’s job; break down starch. It involved the placing of human amylase derived from human saliva and fungal amylase into various test tubes that contained 1% starch solution. The test tubes were then placed into baths of different temperatures and left to settle for five minutes, followed by the adding of the amylases to the starch solutions. Then a few drops of the solutions were then extracted from the tubes placed respectively on a spot plate that was marked for the different temperatures and time. Next, it was mixed with three drops of iodine to determine the extent of starch hydrolysis. This was done a total of 5 times with two-minute intervals in between and the results were recorded accordingly. It was discovered that the optimal temperature for human amylase is roughly between 40 and 60 degrees and the optimal temperature for fungal amylase is higher at about 60 degrees, a higher and narrower range that that of human amylase. How this conclusion was determined was by adding the iodine to the solutions after the two-minute intervals and observing the color change of the iodine to see if the starch has broken down. Iodine was used because when iodine, which is yellow, comes into contact with starch it reacts by darkening and turning dark blue to black. If the starch is still present in the solutions being tested, mixing iodine into them would produce a dark pigment. If hydrolysis occurred, then the iodine would retain its yellow color. Put simply Black =no hydrolysis, Yellow= 100% hydrolysis. Anything in between, such as a brown color, would mean hydrolysis may have occurred but not at 100%, meaning there are still some bonds present in the starch and this would make the temperature at which it is being tested not optimal.

Introduction:
Amylase is an enzyme that breaks down starch and this experiment focused on what is the optimal temperature at which human and fungal amylase. Amylase is found in various organisms and in fact research has shown that there is a larger amount of amylase proteins in the saliva of populations that consume more starch, such as in areas of greater vegetation. By contrast, there is a smaller amount of amylase enzymes in the saliva of populations of arctic conditions. This is an example of natural selection in a positive manner, because it benefits the organism directly (Perry et al., 2007). Amylase turns starch polymers into monomers such as maltose. Therefore, if amylase has done it’s job right, there should be no presence of starch when the amylase is at it’s optimal temperature, this is tested by adding a few drops of iodine to the solution. If the solution turns yellow after adding the iodine, there is no starch present, if it turns dark blue or black, starch is present. This is because of the way iodine reacts to the shape of the starch molecule (Goldina and Simms, 2010). Knowing the optimal conditions for amylase can prove to be very important commercially. Some forms of amylase are used to remove starch chains from sugarcane in sugarcane processing plants. Not providing the optimal temperatures and other conditions can result in an unwanted product and can even lead to conflict between the producers and consumers (Montes et al., 2008). If temperatures are too cold, the molecules themselves move slower and take much longer to react until they finally cease moving all together if temperatures are low enough. Temperatures that are too high, on the other hand, cause the enzyme to denature and thus not allow the reactions to take place between enzyme and molecule because they no longer fit together (Goldina and Simms, 2010). In this experiment it is hypothesized that the amylase/starch solutions will not hydrolyze at 100% in either the ice water bath (0˚) nor in the 95˚ bath. This is hypothesized because human and fungal amylase is being tested, and neither human nor fungus’ thrive in extreme temperatures. Both of these organisms do best in warm temperatures so neither the very cold 0˚ or the very hot 95˚ should provide 100% hydrolysis. There are, however, forms of amylases that function at an optimal level in very cold climates, such as amylase found in Pseudoalteromonas haloplanktis, an arctic bacteria (Khawar et al., 2005).

Materials and Methods:
Various materials were used to conduct this experiment on optimal temperatures for human and fungal amylase. The first obvious materials used were the amylases. The human amylase was derived from human saliva and the fungal amylase from a pre-made solution. A solution of 1% starch was also used as well as 16 test tubes were; four for human amylase, four for fungal amylase, and eight for the starch solutions. Some skin diseases are more than skin deep. Psoriasis is the most prevalent autoimmune disease in the country, affecting as many as 7.5 million Americans. This noncontagious condition often appears on the skin as red, scaly patches that itch and bleed. Psoriasis frequently starts between the ages of 15 and 35, but can appear at any time. New research in the news indicates that chronic inflammatory conditions such as psoriasis may actually cause HDL cholesterol to change its molecular composition and increase the risk of severe heart disease. ?So the reasons to treat Psoriasis are more important now than ever. And because there are so many different medications for the condition, it takes experience and patience for the right treatment or combination of treatments to be successful,? says Dr. Joshua Fox, founder and director of New York & New Jersey based Advanced Dermatology P.C.

As of now, Psoriasis is incurable; however, treatments ranging from topical to systemic can treat it and often clear the symptoms. Psoriasis is marked by an overactive immune system that speeds up the growth cycle of the skin. In normal skin, new cells surface about once a month, but with psoriasis, new cells full with inflammatory cells surface in just three to seven days. Due to this rapid turnover, these cells build up into thick patches that have a silvery, flaking crust. Stress is a common trigger of psoriasis, along with diet, medications, cuts, scrapes, infections and allergies. According to findings presented at the summer 2011 meeting of the American Academy of Dermatology, when psoriasis patients feel stressed about their condition, it can aggravate their symptoms and impact their emotional state, which becomes a vicious cycle adding to their stress. Doctors previously thought that chemicals released by the skin?s nerve endings only stayed in the skin when they were released. But doctors know understand that those chemicals travel to the brain and deplete the chemicals that regulate our emotions, such as serotonin, norepinephrine and dopamine.

Treating Psoriasis

Topical Treatments are usually the first kind of medicine dermatologists use to treat psoriasis. These creams can slow the growth of skin cells and decrease the inflammation of skin lesions.

Phototherapy or light therapy usually involves consistent exposure to ultraviolet light at a doctor’s office or with the sun. It is often used on patients with extensive psoriasis, moderate to severe psoriasis and combined with topical treatments. There are two different kinds of light therapy that utilize either UVA or UVB light.

Ultraviolet light B (UVB)/Ultraviolet light A (UVA) Treatments. Present in sunlight, UVB is an effective treatment for psoriasis because it penetrates the skin and slows the growth of affected skin cells. Treatment involves exposing the skin to an artificial UVB light source for a set length of time on a regular schedule at a doctor?s office. There are two types of UVB treatment, broad band and narrow band. The narrow band UVB releases a smaller range of ultraviolet light. ?Many studies indicate that narrow-band UVB clears psoriasis faster and produces longer remissions than broad-band UVB,? says Dr. Fox. ?It is also more effective with fewer treatments per week than broad-band.? UVA light treatments are combined with a light-sensitizing medication psoralen, which is administered topically or orally. This process, called PUVA, slows excessive skin cell growth and can clear psoriasis symptoms for various amounts of time. However there are more side effects with this treatment, including special precautions for the eyes and greater risk of burning.

Laser Treatments. Laser treatment delivers a single high intensity narrow beam of ultraviolet light of 308 nanometer to target specific smaller areas of skin lesions. It works faster than either the UVB or UVA. It also only treats the affected skin sparing normal skin from side effects. There is no pain or bruising. Patients usually receive two treatments per week until clear.

Pulsed dye laser. Using a dye and different wavelength of light than other UVB-based treatments, pulsed dye lasers destroy the tiny blood vessels that contribute to the formation of psoriasis lesions. Treatment consists of 15- to 30-minute sessions every three to four weeks and it usually takes two to six sessions to clear the target lesion. It unfortunately has bruising and there is some discomfort.

Systemic Treatments are prescription drugs given in the form of a pill or an injection. Because of their potential risks, systemic treatments are mostly used to treat more severe cases of psoriasis which have failed the moderate treatments listed above. These systemic treatments include methotrexate, Acitretin, Cyclosporin, Hydroxyurea and Biologics.

Biologics. Biologics are systemic treatments made from proteins produced by living cells. They block parts of the immune-system process that drives psoriasis. Scientists are excited about a new biologic that may even keep psoriasis at bay: The October 2011 issue of the New England Journal of Medicine chronicles a study wherein German researchers administered an experimental treatment of briakinumab, or a commonly used drug, methotrexate. Briakinumab stems an overactive immune system by blocking two proteins that drive inflammation. This experimental drug reduced psoriasis symptoms by 75 percent in more than 80 percent of the study participants, compared with only 40 percent of people who took methotrexate a long time accepted treatment for psoriasis. After a year follow-up, nearly 60 percent of participants taking briakinumab had no skin lesions, compared with fewer than 20 percent of participants taking methotrexate.

?Patients today have many options for treating psoriasis. We encourage people to talk with their dermatologist to determine the best course of action for their specific case,? adds Dr. Fox.
